摘要:

目的 探讨吸烟影响重度哮喘的机制,为治疗由吸烟引起的重度哮喘奠定理论基础。方法 本文以基本免疫仿真器(basic immune simulator,BIS)为平台,仿真出TH17对免疫系统影响的模型(agent based modeling,ABM),与BIS已仿真出的DCs模型对比,并结合吸烟的临床数据,验证模型的正确性。结果 本文提出吸烟引起重度哮喘可能的机制是:烟雾中的氧化物能激发成熟DCs细胞产生IL-8细胞因子,随着烟雾作用时间的延长,产生的IL-8增多,使吸烟的哮喘者加重病情。结论 DCs—IL23—TH17—IL8—嗜中性炎症可能是导致重度哮喘的路径。以TH17为靶标,减少DCs,切断IL23—TH17—IL8路径,可以控制吸烟引起重度哮喘的症状。

Objective To explore the mechanism of severe asthma caused by smoking and establish the  theoretical basis for the therapy of severe asthma with smoking.Methods We take the basic immune simulator (BIS) as a platform to simulate TH17 models for immune system influence,compare with the DCs models made in BIS,and combine smoking clinical data to verify the models’ correctness.Results The mechanism of severe asthma caused by smoking may be that the oxidants in smoking can stimulate mature DCs to produce IL-8,the longer acting time with smoking,the more IL-8 agents are produced and asthma becomes more severe.Conclusions DCs—IL—23—TH17—IL8- neutrophile inflammation may be the path of severe asthma.TH17 as a target,DCs reduced and pathway of IL23—TH17—IL8 cut off,symptom with severe asthma caused by smoking may be controlled.
