海藻酸钠复合物治疗膝关节软骨缺损的研究进展

摘要:

膝关节是人体主要承重关节,其软骨缺损是组织工程技术中正着力解决的难题。软骨组织中细胞含量
少,主要成分是胶原蛋白和蛋白聚糖,修复能力有限。海藻酸钠因其特殊的固液型可转换特性、良好的生
物相容性,以及与软骨基质类似的结构,而成为修复软骨缺损的重要支架材料。很多实验支持海藻酸钠复
合物支架治疗膝关节软骨缺损。三维培养是软骨组织工程细胞体外培养的重要环境条件。自体软骨细胞、
分化前或未分化的骨髓间充质干细胞和基因转染的细胞等均可获得良好的治疗效果。前期研究结果发现海
藻酸钠通过动态压缩加载、流体混合优化、微流体等有极好的培养效果。在软骨细胞的体外培养或诱导培
养中,要立足于为细胞创造与体内类似的环境。海藻酸钠复合物支架趋于复合化、复杂化,应用上要具体
分析支架的孔隙率、力学强度、生物相容性等各方面数据。

Knee is the main bearing joint,whose cartilage defect is the important problem
in tissue engineering techniques. Cartilage is composed of collagen protein and proteoglycan
and a few cells. So the repair ability of cartilage is limited. Sodium alginate can transfer
between solid and liquid,has good biocompatibility and is similar with cartilage matrix.
Sodium alginate is the important scaffold material in tissue engineering. Many studies show
sodium alginate complex can be used in the treatment of knee-joint cartilage defect. 3D
environment culture is important in cell culture in vitro. Autologous chondrocytes,pre-
differentiated and undifferentiated mesenchymal stem cells and gene-transferred cells and so
forth are the ideal seed cells. Dynamic compressive loading,media mixing and microfluidic
technology achieve good culture effect. We should create environment similar to the original
for the cells. Scaffolds of sodium alginate complex tend to be complicate. We need to
analyze the data of porosity,mechanical strength,biocompatibility and etc.
