纳米银颗粒的细胞毒性作用及机制初探

摘要:

目的 研究纳米银颗粒对L929细胞的细胞毒性作用,并对其细胞毒性机制进行了初步推测,以判断纳米银和传统银离子抗菌剂在抗菌机制上是否存在差异。方法 以纳米银颗粒为试验样品,微米银颗粒为对照样品,将不同浓度(2.5~500μg/mL)的试样、对照样同L929细胞共同培养24h,采用噻唑盐比色[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide,MTT]法测定各组细胞的相对增殖率(relative growth rate,RGR),采用TEM观察细胞的超微结构。结果 当纳米银颗粒浓度在10~100μg/mL剂量范围时,随着浓度的增加,细胞增殖率减少,细胞毒性逐渐增大,呈现剂量-效应相关性。在50~500μg/mL剂量范围内,相同剂量下的纳米银颗粒引起的细胞毒性显著高于微米银颗粒,而且超微结构观察结果显示只有纳米银颗粒能够通过细胞吞噬进入细胞内部,而微米银颗粒不能进入细胞内部。结论 初步推断纳米银颗粒进入细胞内部使其产生细胞毒性是纳米银颗粒细胞毒性的作用机制,这与微米银颗粒差异显著。这也提示,一旦银颗粒尺寸降至纳米尺度,将会产生新的生物效应并对人体健康产生危害,具有潜在的风险,在使用含纳米银颗粒的医疗器械甚至保健品时应持谨慎态度。

Objective To investigate the cytoxicity of silver nanoparticles (SNPs) and presume the mechanism.Methods In this study,SNPs were test samples and silver microparticles (SMPs) were control samples.SNPs or SMPs were cultured with L929 cells at different doses from 2.5μg/ml to 500μg/ml,respectively.After 24 hours of culture,the cells’ relative growth rates (RGRs) were tested with MTT colorimetric assay and the ultrastructures of cells were evaluated with transmission electronic microscopy (TEM).Results The cytoxicity induced by SNPs was more severe than by SMPs at the same dose (50μg/ml-500μg/ml).And a dose-effect could be observed at the SNPs’ dose between 10μg/ml and 100μg/ml (the higher the doses,the lower the RGR,the larger the cytoxicity).SNPs could be phagocytozed by L929 cells and enter into the cells,while the SMPs did not.Conclusions The cytoxicity mechanism of SNPs is different from that of SMPs and the key factor is SNPs could enter into cells yet SMPs could not.Once the dimension of silver particles is reduced to nanoscale,SNPs may have adverse effects on human health.We should be cautious with SNPs-based medical devices in clinical practice.
